Glutathione reductase (GR) is a crucial enzyme in the human body's oxygen reduction system. As a key player in the glutathione redox cycle, it catalyzes the conversion of oxidized glutathione (GSSG) into reduced glutathione (GSH), playing a vital role in maintaining the activity and reducing properties of sulfhydryl proteins, preserving cell integrity, and preventing oxidative degradation of hemoglobin.
Measuring serum glutathione reductase activity can aid in diagnosing liver-related diseases, including hepatitis, cirrhosis, obstructive jaundice, and liver tumors.
Detection Principle of Glutathione Reductase (GR)
Glutathione reductase catalyzes the reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH) using NADP+H as a cofactor. During this process, NADP+H is oxidized to NADP++. NADP+H exhibits a characteristic absorption peak at 340 nm, whereas NADP++ does not absorb at this wavelength. By measuring the rate of absorbance decrease at 340 nm, we can determine the rate of NADP+H dehydrogenation, thereby calculating the activity of glutathione reductase (GR).
